Brenner began his professional career in the early 1920s and in May 1923, he fought Jewish boxer Benny Schwartz in Len's hometown of Lancaster, Pennsylvania. The bout officially ended in a no decision, but the popular verdict favored Schwartz, one of the top flyweight contenders at that time. The following month, the two fighters had a rematch in Lancaster and again fought to a no decision.

In October 1923, Brenner fought Battling Murray, a former contender for the world flyweight championship. The bout, which also took place in Lancaster, ended in a no decision. In April 1925, Len fought Joe Lynch, the former world bantamweight champion, and won an eight round decision. Two weeks after defeating Lynch, Brenner then had a rematch with Benny Schwartz and won an eight round decision. Both fights were in Lancaster.
