Klempner first joined the Penn varsity as a junior in 1932. That year, the Quakers finished with a record of 10-11 (2-8 in the Ivy League) and Lester was the team's second leading scorer in conference play with 42 points (16th in the Ivy League).

In 1933, Klempner was Penn's captain and the team improved to a 12-6 overall record with a 6-4 record in conference play. During the tournament season, Lester scored 33 points, fourth most on the team and 26th in the Ivy League.
